Nicotinic Acetylcholine Receptor Agonist Program (Alzheimer’s Disease & Schizophrenia)

  • EnVivo is developing a robust program of selective, orally active alpha 7 agonists for treatment of the cognitive decline in Alzheimer’s disease and Schizophrenia.
  • The lead compound EVP-6124 has successfully completed clinical Phase Ia testing (SAD & MAD) in healthy young volunteers and Phase Ib/IIa trials in Schizophrenia.
  • Phase Ib/IIa trials of EVP-6124 in Alzheimer patients are ongoing.
  • EVP-4473 is a follow-up compound and has successfully completed preclinical development.  
  • The program was in-licensed from Bayer Healthcare and included an extensive and exclusive patent estate surrounding the compounds’ composition-of-matter.


Histone Deacetylase (HDAC) Program (Alzheimer’s Disease)

  • EnVivo is developing a CNS-penetrant, orally bioavailable first in class histone deactylase inhibitor (HDACi) EVP-0334 that is being pursued for use as a cognition enhancing agent in neurodegenerative diseases.
  • The program has entered clinical trials.
  • EnVivo has an exclusive worldwide license from MethylGene, Inc.(Montreal, Québec; TSX:MYG, www.methylgene.com), to operate in the field of HDAC inhibitors for neurodegenerative diseases.

 

Gamma-Secretase Modulator (GSM) Program (Alzheimer’s Disease)

  • EnVivo is developing a CNS-penetrant, orally bioavailable gamma-secretase modulator for disease modifying therapeutic intervention in Alzheimer’s disease.
  • This approach avoids the potential associated toxicities of general gamma-secretase inhibition (GSI) since it does not affect Notch processing and only lowers the toxic species Abeta42. Total levels of Abeta peptides are not altered.
  • EnVivo’s GSM lead compound EVP-0962 has demonstrated acute Abeta42 lowering in non-transgenic animals and transgenic (TG2576) mice and has entered preclinical development.
