EnVivo is developing a CNS-penetrant, orally bioavailable gamma-secretase modulator for disease modifying therapeutic intervention in Alzheimer’s disease.
This approach avoids the potential associated toxicities of general gamma-secretase inhibition (GSI) since it does not affect Notch processing and only lowers the toxic species Abeta42. Total levels of Abeta peptides are not altered.
EnVivo’s GSM lead compound EVP-0962 has demonstrated acute Abeta42 lowering in non-transgenic animals and transgenic (TG2576) mice and has entered preclinical development.
